<div>The therapeutic potential of mRNA is extensively discussed in times of search for vaccines in the research of COVID-19. The idea is to provide the human body with mRNA, encoding therapeutic proteins, which the immune system uses to fight the virus.<sup>1</sup></div>

<div>RNA can be produced and applied much easier compared to the full-length therapeutic proteins it is encoding for. In order to generate pure mRNA to administer, it was found that purification by chromatographic means is highly important.<sup>2</sup></div>

It is highly important that the chromatography is carried out at elevated temperatures in order to denature the RNA itself and hence simplify its structural diversity and yield a successful purification. It consists of a ternary low pressure system pump, which not only gives you a reliable formation of binary gradient between water including 1.0 M TEAA (pH 7) and acetonitrile, but also allows the direct sample application of large volume sample onto the column by the system pump via its separate sample inlet port. The flow rate range of this pump lies between 10 to 80 ml/min. Sample Injection via a sample- or Variloop is possible with the integrated injection valve. After the injection valve solvent as well as sample will be heated by an eluent heater and then applied onto the column, which additionally is heated by a heating sleeve to ensure perfect thermal purification conditions. Product identification is realized by the variable single wavelength detector at 260 nm or the wavelength of your choosing. After purification fractionation is carried out by the fractionation valve.</div>

<p align="justify">The AZURA Pilot Prep LC 1000 HPG system is a complete binary high pressure gradient (HPG) HPLC system for the purification of samples in the laboratory scale. It is designed and optimized for the purification of gram samples. This system proposal consists of two preparative HPLC pumps, a UV/VIS detector and a freely customizable combination module. The AZURA P 2.1L pump covers a wide flow rate range and pressure capabilities. In the current setup, it supports max. pressures up to 75 bar at flow rates up to 350 ml/min (50 bar from 600 &#8211; 1000 ml/min). The integrated automatic recognition of the pump head with RFID technology allows fast adaptions of the pump for various applications. The HPG setup enables the precise formation of binary gradients. Besides, ternary and quaternary gradients are accessible by integration of additional preparative pumps. The AZURA UVD 2.1L represents a UV/VIS detector with a variable single wavelength for the detection of fast eluting peaks. The system is completed by an AZURA assistant ASM 2.1L for preparative HPLC. The assistant module can be customized to individual requirements. For example, large sample volumes can be applied automatically via the integrated injection valve and feed pump. The AZURA Preparative HPLC system comes with an eluent tray for safe storage of eluent bottles and an integrated leak management system with sensors. Columns (20 mm and 30 mm ID) can be attached with an optional AZURA mounting bracket at both sides of each AZURA element. In the current setup, it supports max. pressures up to 100 bar at flow rates up to 500 ml/min. The integrated automatic recognition of the pump head with RFID technology allows fast adaptions of the pump for various applications. The LPG valve block enables the accurate formation of binary or binary gradients with a maximum flow rate of 500 ml/min. The system is completed by an AZURA assistant ASM 2.1L for preparative HPLC. This assistant includes the detector UVD 2.1S, a feed pump and a 12 port multipositions valve for fractionation. The AZURA HPLC system comes with an eluent tray for safe storage of eluent bottles and an integrated leak management system with sensors. Columns (20 mm and 30 mm ID) can be attached with an optional AZURA mounting bracket at both sides of each AZURA element. In the current setup, it supports max. pressures up to 225 bar at flow rates up to 100 ml/min (200 bar from 150 &#8211; 250 ml/min). The integrated automatic recognition of the pump head with RFID technology allows fast adaptions of the pump for various applications. The HPG setup enables the precise formation of binary gradients. Besides, ternary and quaternary gradients are accessible by integration of additional preparative pumps. The AZURA UVD 2.1L represents a UV/VIS detector with a variable single wavelength for the detection of fast eluting peaks. The system is completed by an AZURA assistant ASM 2.1L for preparative HPLC. The assistant module can be customized to individual requirements. For example, to enable automatic injection and fraction collection, the assistant module contains three integrated modules:</p>
<ul>
<li>Large sample volumes can be applied automatically via the integrated injection valve and feed pump.</li>
<li>The integrated fractionation valve enables sample fractionation without an additional fraction collector.</li>
<li>Under isocratic conditions, the consumption of solvent can be reduced by the use of solvent recycling at high flow rates. Peak recycling allows improving the resolution of a pair of closely eluting solutes (e.g., a pair of enantiomers) by automated repeated processing of the peaks.</li>

<p align="justify">The AZURA Pilot Prep LC 250 LPG system is a complete low pressure gradient (LPG) HPLC system for the purification of samples in the laboratory scale. The AZURA Preparative HPLC system is designed and optimized for the purification of milligram to gram samples. The system proposal consists of a preparative HPLC pump with ternary LPG valve block and an Assistent ASM 2.1L. Large sample volumes can be introduced via the third LPG channel. For smal volume size the sample can be injected with a manuel injection valve. The AZURA P 2.1L pump covers a wide flow rate range and pressure capabilities. In the current setup, it supports max. pressures up to 225 bar at flow rates up to 100 ml/min (200 bar from 100 &#8211; 220 ml/min). The integrated automatic recognition of the pump head with RFID technology allows fast adaptions of the pump for various applications. The LPG valve block enables the accurate formation of binary or ternary gradients with a maximum flow rate of 220 ml/min. The system is completed by an AZURA assistant ASM 2.1L for preparative HPLC. This assistant includes the detector UVD 2.1S, an injection valve and a 12 port multipositions valve for fractionation. The AZURA HPLC system comes with an eluent tray for safe storage of eluent bottles and an integrated leak management system with sensors. Columns (20 mm and 30 mm ID) can be attached with an optional AZURA mounting bracket at both sides of each AZURA element. A leak sensor and the capillary guidance as well as the optional Mobile Control with touch screen facilitate the safe and user friendly operation. Modern design with completely demountable fronts turn the system into a visual highlight.</p>

<p align="justify">The AZURA Pilot Prep LC 250 Iso system is a complete isocratic HPLC system for the purification of samples in the laboratory scale. This system proposal is designed and optimized for the purification of milligram to gram samples. It consists of a preparative HPLC pumphttp://www.knauer.net/typo3/clear.gif, a UV/VIS detector and a freely customizable combination module. The AZURA P 2.1L pump covers a wide flow rate range and pressure capabilities. In the current setup, it supports max. pressures up to 225 bar at flow rates up to 100 ml/min (200 bar from 150 &#8211; 250 ml/min). The integrated automatic recognition of the pump head with RFID technology allows fast adaptions of the pump for various applications. The AZURA UVD 2.1L represents a UV/VIS detector with a variable single wavelength for the detection of fast eluting peaks. The system is completed by an AZURA assistant ASM 2.1L for preparative HPLC. The assistant module can be customized to individual requirements. For example, to enable automatic injection and fraction collection, the assistant module contains three integrated modules:</p>
<ul>
<li>Large sample volumes can be applied automatically via the integrated injection valve and feed pump.</li>
<li>The integrated fractionation valve enables sample fractionation without an additional fraction collector.</li>
<li>Under isocratic conditions, the consumption of solvent can be reduced by the use of solvent recycling at high flow rates. Peak recycling allows improving the resolution of a pair of closely eluting solutes (e.g., a pair of enantiomers) by automated repeated processing of the peaks.</li>

<p align="justify">The AZURA Pilot Prep LC 100 HPG system is a complete binary high pressure gradient (HPG) HPLC system for the purification of samples in the laboratory scale. It is designed and optimized for the purification of milligram to few gram samples. This system proposal consists of two preparative HPLC pumps, a UV/VIS detector and a freely customizable combination module. The AZURA P 2.1L pump covers a wide flow rate range and pressure capabilities. In the current setup, it supports max. pressures up to 400 bar at flow rates up to 100 ml/min. The integrated automatic recognition of the pump head with RFID technology allows fast adaptions of the pump for various applications. The HPG setup enables the precise formation of binary gradients. Besides, ternary and quaternary gradients are accessible by integration of additional preparative pumps. The AZURA UVD 2.1L represents a UV/VIS detector with a variable single wavelength for the detection of fast eluting peaks. The system is completed by an AZURA assistant ASM 2.1L for preparative HPLC. <p align="justify">The AZURA® Pilot LC 100 System offers the best conditions for transferring separations from the analytical to the preparative scale. Whether it is simple analysis or purification with 30 mm diameter columns, the Pilot LC 100 System has been designed for a wide range of requirements.</p>
<p><strong>System components</strong></p>
<p align="justify">The system features an AZURA® P 2.1L pump with a maximum flow rate of 100 ml/min. At this flow rate it is possible to purify large sample sizes up to 70 g per day. The injection is carried out via the automatic injection valve installed in the assistant. Thanks to the KNAUER VariLoop, up to 40 ml sample solution can be injected at once or step by step without any dilution. The system is completed by a UV detector and a 16-port fractionation valve, which are also housed in the assistant. This compact and powerful system is controlled by our user-friendly software PurityChrom®. The low pressure gradient module (NDG module) allows operation in both ternary and binary modes. In binary mode, samples can be fed directly to the column via one channel of the NDG module. This allows the system pump to be used simultaneously as a sample pump and very large sample quantities can be applied in an automated manner without any problems.</p>
<p><strong>Injection with VariLoop</strong></p>
<p align="justify">The KNAUER VariLoop is the optimal solution if medium to large sample sizes are to be applied to the column. The VariLoop can be filled completely or partially, as well as completely or partially emptied. This allows very flexible and automated work and the easy exchange between different sample volumes. A constant and reproducible result is achieved for each sample volume. In contrast to all other methods for the injection of large sample volumes, the</p>
<p align="justify">KNAUER VariLoop allows a nearly lossless sample application.</p>

<p align="justify">The AZURA Pilot Prep LC 100 LPG system is a low pressure gradient (LPG) HPLC system for the purification of samples in the laboratory scale. The AZURA Preparative HPLC system is designed and optimized for the purification of milligram to gram samples. The system proposal consists of a preparative HPLC pump with ternary LPG valve block and an Assistant ASM 2.1 L with sample pump for large sample volums, UV-detector UVD 2.1S and a fractionation valve. Smal sample size can be injected with the manual injection valve. The AZURA P 2.1L pump covers a wide flow rate range and pressure capabilities. In the current setup, it supports max. pressures up to 400 bar at flow rates up to 100 ml/min. The integrated automatic recognition of the pump head with RFID technology allows fast adaptions of the pump for various applications. The LPG valve block enables the accurate formation of binary or ternary gradients. The system is completed by an AZURA assistant ASM 2.1L for preparative HPLC. This assistant includes the detector UVD 2.1S, an additional feed pump and a 16 port multipositions valve for fractionation. The AZURA Preparative HPLC system comes with an eluent tray for safe storage of eluent bottles and an integrated leak management system with sensors. Columns (20 mm and 30 mm ID) can be attached with an optional AZURA mounting bracket at both sides of each AZURA element. A leak sensor and the capillary guidance as well as the optional Mobile Control with touch screen facilitate the safe and user friendly operation. Modern design with completely demountable fronts turn the system into a visual highlight.</p>

Fractions can be collected into 96 well microplates, standard tube sizes, and bottles. For essentially unlimited volumes, funnel racks can direct fluids to any collection vessel or downstream process. The AZURA Preparative HPLC system comes with an eluent tray for safe storage of eluent bottles and an integrated leak management system with sensors.</p>
<p align="justify">Columns (20 mm and 30 mm ID) can be attached with an optional AZURA mounting bracket at both sides of each AZURA element. It is designed and optimized for the purification of milligram to gram samples. The system proposal consists of a preparative HPLC pump, a UV/VIS detector and a freely customizable combination module. The AZURA P 2.1L pump covers a wide flow rate range and pressure capabilities. In the current setup, it supports max. pressures up to 400 bar at flow rates up to 100 ml/min. The integrated automatic recognition of the pump head with RFID technology allows fast adaptions of the pump for various applications. The AZURA UVD 2.1L represents a UV/VIS detector with a variable single wavelength for the detection of fast eluting peaks. The system is completed by an</p>
<p align="justify">AZURA Assistant ASM 2.1L for preparative HPLC. The assistant module can be customized to individual requirements. For example, to enable automatic injection and fraction collection, the assistant module contains three integrated modules:</p>
<ul>
<li>Large sample volumes can be applied automatically via the integrated injection valve and feed pump.</li>
<li>The integrated fractionation valve enables sample fractionation without an additional fraction collector.</li>
<li>Under isocratic conditions, the consumption of solvent can be reduced by the use of solvent recycling at high flow rates. Peak recycling allows improving the resolution of a pair of closely eluting solutes (e.g., a pair of enantiomers) by automated repeated processing of the peaks.</li>

<p align="justify">The AZURA Preparative HPLC system comes with an eluent tray for safe storage of eluent bottles and an integrated leak management system with sensors. 										<content:encoded><![CDATA[<p align="justify">Automated binary semipreparative system with a maximum flow rate of 50 ml/min</p>
<p align="justify">The AZURA Lab Prep LC 50 HPG system with 50 ml pump head is a completely automated semi-preparative high pressure gradient HPLC system. The system consists of an Autosampler 6.1L, Pump P 6.1L, a Detector UVD 2.1L, Fraction Collector Foxy R2 and comes with an eluent tray for safe storage of up to six bottles. The Autosampler 6.1L can inject from up to 768 positions when equipped with microtiter plates (either high or low formats) or from up to 108 standard 2 ml sample vials. The pump can deliver flow in the range of 0.01 – 50 mL/min at pressures up to 300 bar. The AZURA Binary Pump contains two identical high pressure pumps (300 bar), 2 × 2-channel inlet solvent selection valve and the newly developed AZURA mixer, a low-volume mixing device. The Foxy R1 fraction collector is ideal for applications including prep HPLC, protein, and peptide purifications. The column can be attached anywhere to the system via the magnetic clip. Solvent bottles can be stored in an eluent tray on top of the system. A leak sensor, capillary guides as well as a tablet with Mobile Control are features rarely found in other preparative HPLC systems. Modern design with completely demountable fronts turn the system into a visual highlight. The stackable elements of AZURA allow to create complex system solutions requiring only a minimum of space.</p>

<p align="justify">This very compact system is specially designed to separate different natural components, like carbohydrates, alcohols and organic acids. It is optimized for the separation of several hundred mg to gram per day.</p>
<p><strong>System components</strong></p>
<p align="justify">The heart of the system is the superior AZURA refractive index detector RID 2.1L HighFlow with an outstanding linear range and sensitivity. The intelligently designed optical unit with advanced temperature control ensures high sensitivity, fast baseline stabilization, and excellent reproducibility. The wide linear dynamic range and 100 ml/min maximum flow rate make the AZURA RID 2.1L HighFlow the perfect choice for most laboratory tasks and it is an excellent device for semi-preparative purification of sugars using the KNAUER Eurokat columns. The other systems component is the AZURA Assistent ASM 2.1L. This sugar purification assistent combines the functionality of two different devices, a pump and a valve. The isocratic AZURA Pump P 4.1S can work with flow rates up to 50 ml/min and a maximum pressure of 150 bar. The 12 port multi-position valve AWA20BG for fractionation is made of stainless steel and has a maximum flow rate of 500 ml/min. This price attractive system is made in Germany and fulfills the highest quality standards. <p align="justify">The AZURA Compact Prep LC 50 iso System with 50 ml pump head is a complete semi-preparative high pressure HPLC system. Due to its extremely compact dimensions, the system fits even onto the smallest laboratory bench. The System consists of an Assistant ASM 2.1L with integrated pump, detector and fraction collector valve and comes with an eluent tray for safe storage of up to six bottles. Injection is performed via a manual injecton valve, which is fixed to the side of the detector. The column can be attached anywhere to the system via magnetic clip. Solvent bottles can be stored in an eluent tray on top of the system. A leak sensor and the capillary guidance as well as a tablet with Mobile Control. Modern design with completely demountable fronts turn the system into a visual highlight. The stackable elements of AZURA allow to create complex system solutions requiring only a minimum of space.</p>
